The Collegio Urbino has the faculty of doctoring, as many documents prove, and immediately acquires a good reputation. Its prerogatives are extended by the bull of Pius IV Sedes Apostolica Gratiarum Abundantissima Mater of February 22, 1564, which confers on the College of Doctors wider prerogatives "ad instar nonnullorum aliorum Italiae Collegiorum".

The origin of the University of Urbino is closely linked to the history of the homonymous Duchy which, at the beginning of the 16th century, was Vicariate of the Holy Roman Church, governed in succession by Guidubaldo I da Montefeltro (1482-1508) and by Francesco Maria I Della Rovere (1508-1538).
